The latest guest villain. But I wonder how this sort of thing will play with TV viewers who know who Supergirl and the other principals are, but may not have even heard of the sundry comic book characters..

I think a lot of viewers will vaguely remember these villains from the old comics. But for those that don't, the hooks that were set in the Pilot should suffice.
The crash of the Phantom Zone prison dumped a large quantity of dangerous escaped convicts on Earth, all of them aliens of one kind of another. In the show, the general public doesn't know that, but the viewers do. So it makes sense that these people keep showing up.
The trick is to avoid the random "bad guy of the week" thing and show them as part of a larger story arc. That will be assured by the arrival of the General, who will harness them or release them as she wishes to get her revenge on Kara. Hopefully that conflict will grow more complex with time, with Kara and the General meeting. Lets see... two Kryptonian superwomen duking it out. Who wants to see that?
What hasn't been said so far is that given the General is an El family member, and she wants to kill the daughter of Zor El, her niece, one would think she has a beef with the son of Jor El as well. Or is this a bit of family politics that doesn't involve the more dominant Jor El side of the family? Wonder if they will mention any of that. It's a wonderful opportunity for the cousins to work together, but I expect that's out of the question for Season 1. After Batman v Superman has run its course in the theaters, perhaps more things become possible. Or less. Time will tell.

Point of correction:
The Generel is NOT of the House of El, and if you look at the video she's not wearing the House of El Logo on her uniform (It's not easily visible, but I took a screen shot and played with the contrast to be sure). She's Kara's Aunt (her mother's sister). Kara's father Zor-El is of the House of El. In this it works much as it does in the US ,she has her Dad's last name and house, not her mothers.
Supergirl is of the General's bloodline, but Kal-El isn't related to her at all biologically. She's his Uncle's wife's sister? Or easier, she's his Aunt's Sister, but not his Aunt. She's Supergirl Aunt.

You are good. Really good. "She's his Uncle's wife's sister". I would never have figured that out.
I've been easily confused since someone claimed to be my "3nd cousin once removed" or some such thing. <exploding head>
She's not an El. Got it. But she's Kryptonian. So the fight will still be awesome.

When you just think about Supergirl, it's easy "Aunt".
She's also a twin for Kara's mother, so it makes the relationship a bit more personal. And I'd be surprised if that wasn't brought up. If you were someone who lost her mother at a young age (and her whole planet) it'll be heartbreaking to meet her twin, and find her evil and someone you must fight.
A question is if Kara is aware that her mother has an identical twin. It's possible that she's been in the Phantom Zone since before Kara was born.
Considering Kara seems to be unaware of what her mother really did, I wouldn't put this out of consideration.
